Blocked on penetration

It cannot reach the tissue

A molecule that stalls at the tissue barrier, or reaches the target only through a destructive or invasive procedure. This is the case with human proof of concept behind it.

Blocked on stability

It cannot survive the format

An active that forces a cold chain, shortens shelf life, or will not hold in the product format the market wants.

Blocked on exposure

It works, but the dose is capped

Tolerability sets the ceiling before the biology does. Even a molecule that reaches the target can still be impossible to keep where it was put, leading to off target side effects.
